ABSTRACT Advancing microbiome–gut–brain axis science requires systematic, rational and translational approaches to bridge the critical knowledge gaps currently preventing full exploitation of gut microbiome as a tractable therapeutic target for gastrointestinal, mental brain health. Current research is still marked by many open questions that undermine widespread application humans. For example, lack mechanistic understanding probiotic effects means it remains unclear why even apparently closely related strains exhibit different in vivo. live microbial psychobiotics, consensus on their adjunct treatments conventional neuromodulators, use unmedicated populations or at‐risk cohorts with sub‐clinical symptomatology warranted. This missing information both sides equation when treating central nervous system (CNS) conditions makes psychobiotic challenging, especially compared other pharmaceutical functional food approaches. Expediting transition from positive preclinical data proven benefits humans includes interpreting promises pitfalls animal behavioural assays, well navigating mechanism‐informed decision making select right microbe(s) job. In this review, we consider how these decisions can be supported light accrued range clinical studies across healthy, pathological study populations, where specific have been evaluated context gastrointestinal physiology, function behaviour. Examples successful, partial unsuccessful translation bench bedside are considered. We also discuss developments silico analyses enhanced our moved towards pinpointing host–microbe interactions most important optimal gut–brain function. Combining assays vitro ex vivo domains incorporating model organisms prime discovery pipelines promising rationally selected candidates.

The maternal microbiome is emerging as an important factor that influences the neurological health of mothers and their children. Recent studies highlight how microbial communities in gut can shape early-life development ways inform long-term trajectories. Research on neurodevelopmental effects microbiomes expanding our understanding microbiome-gut-brain axis to include signaling across maternal-offspring unit during perinatal period. In this Review, we synthesize existing literature modulates brain function behavior both developing offspring. We present evidence from human animal showing interacts with environmental factors impact risk for abnormalities. further discuss molecular cellular mechanisms facilitate crosstalk neuromodulation. Finally, consider advancing these complex interactions could lead microbiome-based interventions promoting offspring health.

Background: Emerging evidence suggests that the maternal microbiome plays a crucial role in shaping fetal neurodevelopment, immune programming, and metabolic health. Dysbiosis during pregnancy—whether gastrointestinal, oral, or vaginal—can significantly influence pregnancy outcomes long-term child Materials Methods: The search was performed using databases such as PubMed, Scopus, Google Scholar including research published from January 2000 to 2025. keywords used were “Fetal Programming”, “ Maternal Immune Activation”, “Maternal microbiome”, “Microbiota–Gut–Brain Axis”, “Pregnancy Dysbiosis”. Results: undergoes substantial changes pregnancy, with alterations microbial diversity function linked conditions gestational diabetes, obesity, preeclampsia. Pregnancy-related dysbiosis has been associated adverse neurodevelopmental outcomes, an increased risk of autism spectrum disorder (ASD), attention-deficit/hyperactivity (ADHD), cognitive impairments offspring. Conclusions: Understanding intricate relationship between microbiota health is essential for developing targeted interventions. Leucine-rich repeat transmembrane proteins (LRRTMs) are synaptic adhesion that regulate synapse development and function. They interact transsynaptically with presynaptic binding partners to promote differentiation. Polymorphisms of LRRTM4, one the four members this protein family, have been linked multiple neuropsychiatric disorders childhood aggression, but underlying mechanisms physiological function LRRTM4 during behavior currently unclear. To characterize role gene for brain function, we combined a battery behavioral assays transcriptomic metabolomic analyses, using zebrafish as model system. Our findings revealed lrrtm4l1, brain-specific orthologue human exhibits region-specific expression pattern similar humans, strong in dorsal telencephalon, area critical regulating emotional-affective social behavior. lrrtm4l1-/- displayed heightened anxiety reduced while locomotion remained unaffected by knockout. Transcriptomic analysis telencephalon identified over 100 differentially expressed genes between wild-type mutant an enrichment pathways related plasticity neuronal signaling. The metabolome showed alterations, particularly dopaminergic adenosinergic neurotransmitter systems. These suggest LRRTMs may functions beyond their established excitatory development, such regulation neurotransmission Targeting therapeutically thus be interesting novel approach alleviate excessive aggression or associated number conditions.

Background/Objectives: Lacticaseibacillus rhamnosus HN001 (HN001) is a probiotic strain widely studied for its potential to improve human health. Previous studies have demonstrated promising results in the improvement of mental well-being, particularly terms increased happiness and support stress management healthy adults. Methods: To further explore these findings, double-blind, placebo-controlled trial was conducted with 120 participants aged ≥ 18 years mild high measured by Perceived Stress Scale (PSS). The were randomly assigned receive either or placebo 28 days. Psychological assessments, including Oxford Happiness Questionnaire (OHQ), completed at baseline, day 14, 28. Secondary outcomes included changes PSS scores, as well depression, anxiety, stress, total score levels DASS-21 questionnaire. Results: While not statistically significant, who received showed an OHQ (mean change, 13.3) scores −8.1) over time compared group 10.2 −6.6, respectively). Furthermore, 39% moved from happy happy, only 29% group. Post-hoc analysis significant interaction between intervention study p-values 0.014 0.043, respectively. No adverse effects observed.
