Abstract Radioresistance is a major challenge in tumor radiotherapy and involves mixture of cellular events, including ferroptosis, new type programmed cell death characterized by the excess accumulation iron-dependent lipid peroxides. In present study, we observed that surviving cancer tissues cells after had significantly greater glutathione to oxidized (GSH/GSSG) ratios lower reactive oxygen species (ROS) malondialdehyde (MDA) levels than nonirradiated tumors cells. Untargeted lipidomic analyses revealed oleic acid (OA) palmitoleic (POA) were most upregulated unsaturated fatty acids irradiated compared with those control IR. Both OA POA could protect from killing effects ferroptosis inducer erastin RSL3, stronger protective effect POA, resulting ROS production POA. Mechanistically, protected caused polyunsaturated acid-containing phospholipids an ACSL3-dependent manner. A mouse model demonstrated ACSL3 knockdown combined imidazole ketone synergistically enhanced antitumor radiation-resistant vivo. Our study reveals previously undiscovered associations between radiation metabolism providing novel treatment strategy for overcoming radioresistance.

Abstract Our previous research has established that hydrogen sulfide (H 2 S) exerts an antagonistic effect against the hippocampal neurotoxicity induced by Rotenone (ROT). However, underlying mechanisms are so far poorly understood. Substantial evidence corroborates involvement of ferroptosis in ROT-induced neurotoxicity. To elucidate protective mechanism H S neurotoxicity, this study explores its regulatory role and mechanisms. We used Fluoro-Jade B staining to detect dead neurons. The levels ferrous ions glutathione (GSH) were measured a kit. ferroptosis-related proteins, including light-chain subunit (xCT), GSH peroxidase 4(GPX4), marker acyl-CoA synthetase long-chain family member 4(ACSL4), ferritinophagy-related protein, ferritin heavy chain 1 (FTH1), sequestosome (p62), ferritinophagy markers autophagosome I/II (LC3I/II), nuclear receptor coactivator 4 (NCOA4), Western blot. findings indicate reduces neuron deaths ROT-exposed rats. Meanwhile, reverses downregulations xCT GPX4, upregulations ion ACSL4 hippocampus ROT. Furthermore, LC3I/II NCOA4, P62 FTH1. Based on these findings, we concluded neuronal death involves inhibiting triggered ferritinophagy.

Cancer is the main leading cause of death worldwide and poses a great threat to human life health. Although pharmacological treatment with chemotherapy immunotherapy therapeutic strategy for cancer patients, there are still many shortcomings during such as incomplete killing cells development drug resistance. Emerging evidence indicates promise inducing ferroptosis treatment, particularly eliminating aggressive malignancies that resistant conventional therapies. This review covers recent advances in important regulatory targets metabolic pathway inducers (focusing mainly on last 3 years) delineate their design, mechanisms action, anticancer applications. To date, compounds, including inhibitors, degraders, active molecules from traditional Chinese medicine, have been demonstrated ferroptosis-inducing activity by targeting different biomolecules pathway. However, strictly defined not yet approved clinical use; therefore, discovery new highly active, less toxic, selective compounds remains goal further research coming years.
