Background The bioactive compound 2-Dodecyl-6-methoxycyclohexa-2,5-diene-1,4-dione (DMDD), derived from the horn root of star fruit, exhibits therapeutic promise through its modulation TGF-β1 pathway and regulation bile acids. Methods In this study, a liver fibrosis model was established in Kunming mice (KM) induced by carbon tetrachloride (CCL4), DMDD (50 mg/kg) administered intragastrically. HE staining, Masson Sirius staining were used to evaluate effect on fibrosis. Illumina sequencing platform detect intestinal flora transcriptome information mouse feces, high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) technology acid content changes feces. Results results show that can mitigate fibrosis-induced damage mice, potentially suppression TGF-β/Smad signaling pathway. Furthermore, increased abundance Lactobacillus , Bacteroides Ruminococcaceae Ruminococcus Oscillospira thereby addressing disturbances regulating metabolism. Conclusion Our study suggests alleviates inhibiting pathway, restoring gut microbiota homeostasis, balancing

The therapeutic effect of immune checkpoint inhibitors (ICIs) in triple-negative breast cancer (TNBC) is unsatisfactory. "cold" microenvironment caused by tumor-associated fibroblasts (TAFs) has an adverse on the antitumor response. Therefore, this study, mixed cell membrane-coated porous magnetic nanoparticles (PMNPs) were constructed to deliver salvianolic acid B (SAB) induce response, facilitating transition from a "hot" tumor and ultimately enhancing efficacy inhibitors. PMNP-SAB, which based coating red blood membrane TAF (named PMNP-SAB@RTM), can simultaneously achieve dual effects "immune escape" "homologous targeting". Under influence external field (MF), SAB be targeted concentrated at site. released tumors effectively inhibit production extracellular matrix (ECM) TAFs, promote T-cell infiltration, responses. Ultimately, combination PMNP-SAB@RTM BMS-1 (PD-1/PD-L1 inhibitor 1) inhibited growth. Finally, study presents precise effective new strategy for TNBC immunotherapy basis differentiation microenvironments.

Liver fibrosis is a globally prevalent chronic liver disease, often representing the advanced stage of various conditions. Despite its widespread occurrence, there currently no widely accepted or effective treatment for fibrosis. However, increasing evidence supports efficacy Traditional Chinese Medicine (TCM) in inhibiting progression In this study, we explored effects and potential mechanisms Qizhu-Ruogan-Granules (QZRG), formulation from Affiliated Hospital Chengdu University TCM, on carbon tetrachloride (CCl4)-induced mice. A total 40 male C57BL/6J mice were randomly divided into five groups (n = 8 per group), with induced by injecting 10% CCl4 15 weeks. From 7th week onward, QZRG granules administered orally to at low, medium, high doses. To assess function, serum levels alanine aminotransferase (ALT), aspartate (AST), alkaline phosphatase (ALP) measured. morphology evaluated using hematoxylin-eosin (H&E) Masson's trichrome staining, while gene protein expression analyzed through quantitative reverse transcription polymerase chain reaction (RT-PCR) western blot techniques. The results showed that significantly reduced AST, ALT, ALP CCl4-treated mice, alleviated damage, collagen accumulation. Furthermore, inhibited apoptosis-related proteins BAX, Caspase9, Caspase8, Caspase3, reducing P2Y14 fibrotic tissues. Additionally, suppressed proliferation activated hepatic stellate cells. Our findings suggest may exert anti-fibrotic downregulating effectively slowing
