Abstract Ferroptosis is an important pathological process in benign prostatic hyperplasia (BPH) that can exacerbate lower urinary tract symptoms (LUTS). Hydroxycitric acid (HCA), active component of the traditional Chinese medicine Garcinia cambogia, exhibits anti-inflammatory and anti-tumor properties, preclinical studies have confirmed its anti-obesity effects. In this study, we investigated effects HCA on propionate testosterone-induced BPH found treatment significantly reduced ferroptosis prostate. Further mechanistic indicated Nrf2 crucial for HCA-induced upregulation GPX4, which mitigates by reducing iron accumulation lipid peroxidation. Overall, these findings provide insights into pathogenesis BPH.

Ferroptosis is a novel form of regulated cell death characterized by the iron-dependent accumulation lipid peroxides. Recent research has suggested that ferroptosis in osteoblasts contributes to steroid-induced osteonecrosis femoral head (SONFH). However, relationship between and SONFH remains unclear. In this study, vitro experiments show dexamethasone (Dex) treatment reduces expressions key regulators, SLC7A11 GPX4, MC3T3-E1 cells. This reduction leads decrease intracellular glutathione (GSH) levels, accompanied elevated levels total iron, malondialdehyde (MDA), reactive oxygen species (ROS). Importantly, inhibitor ferrostatin-1 (Fer-1) effectively reverses Dex-induced Furthermore, RNA-seq analysis reveals long noncoding RNA (lncRNA) XR_877193.1is significantly upregulated Dex-treated Functional studies demonstrate knockdown lncRNA XR_877193.1 promotes osteogenic differentiation inhibiting cells, whereas its overexpression exacerbates via ferroptosis. Kyoto Encyclopedia Genes Genomes (KEGG) enrichment differentially expressed enriched ferroptosis-related pathways, including PI3K/AKT signaling pathway. Moreover, inhibitors reverse cells inhibited knockdown. Collectively, our findings indicate exerts anti-ferroptosis effects stimulating pathway, suggesting promising therapeutic strategy for attenuating SONFH.

Abstract This study aimed to evaluate the effectiveness of acupuncture for benign prostatic hyperplasia (BPH). We searched PubMed, EMBASE, Cochrane Library, Chinese Biomedical Literature Database, National Knowledge Infrastructure, Wanfang Data, and China Science Technology Journal Database from their inceptions February 1, 2022. The language was restricted English Chinese. Two researchers independently screened literature, extracted data, evaluated risk bias included according Handbook 5.1.0. A meta-analysis performed using Review Manager version 5.3. Twenty-two studies with 1,765 participants were selected be incorporated in meta-analysis. experimental groups (EGs) showed greater benefit on therapeutic effects (relative [RR]: 1.23, 95% CI: 1.16, 1.3; P < 0.00001), International Prostate Score Scale (mean difference [MD]: −2.06, −3.17, −0.96; = 0.0002), maximum urinary flow rate (MD: 1.7, 0.89, 2.52; 0.0001), postvoid residual urine volume −8.25, −12.14, −4.36; quality life −0.55, −0.8, −0.29; p 0.0001) compared control groups, whereas prostate −0.87, −2.66, 0.92; 0.34) not significantly improved EGs. Nevertheless, no statistical variation reduction adverse reactions observed. Acupuncture therapy, a wildly applied complementary–alternative treatment, may help management BPH.

Benign prostatic hyperplasia (BPH) and prostate cancer (PCa) are prevalent diseases in aging male populations, with significant implications for quality of life healthcare burden. While cardiovascular health (CVH) lifestyle factors have been linked to chronic diseases, their association remains underexplored. This study investigates the relationship between CVH, assessed using Life's Crucial 9 (LC9) score, risk including BPH PCa, a large UK cohort. Data from 26,656 participants Biobank were analyzed. The LC9 an expanded CVH metric incorporating mental health, was calculated based on physical, metabolic, psychological factors. Logistic regression restricted cubic spline analyses performed examine associations score diseases. Subgroup interaction conducted explore potential modifiers. A higher significantly associated lower both continuous categorical models (P < 0.001). demonstrated linear dose-response relationship, inflection point at 72.5. analysis revealed stronger protective effects socioeconomic deprivation. However, no observed PCa risk, even subgroup analyses. highlights importance overall reducing emphasizing need sustained healthy behaviors. absence link suggests distinct pathophysiological mechanisms warrants further research. These findings provide valuable insights targeted prevention management strategies health.

Benign prostatic hyperplasia (BPH) poses a significant public health challenge, affecting substantial portion of aging men worldwide. Current therapeutic options offer limited efficacy. The pathogenesis BPH is multifactorial, involving ferroptosis, oxidative stress, and chronic inflammation. Hydroxycitric acid (HCA) natural compound with diverse pharmacological activities, including the inhibition anti-inflammatory, anti-oxidative anti-tumor effects. However, its role in remains unexplored. This study aimed to investigate effects HCA on elucidate underlying mechanisms, goal providing novel insights for treatment. C57BL/6J mice were used establish model induced by testosterone propionate (TP). Animals then randomly assigned following groups: Sham, BPH, + Lip-1, Bru, HCA. Prostate index (PI) was determined, histopathological changes evaluated hematoxylin eosin (HE) staining. Mitochondrial morphology analyzed TEM. levels Fe2+, MDA, GSH prostate tissues measured. Western blot analysis performed assess protein expression Nrf2 GPX4. Compared Sham group, group exhibited typical features hyperplasia, epithelial cell proliferation, increased glandular lumen size. Concurrently, ferroptosis markers Fe2+ (P < 0.01) MDA 0.001) significantly elevated, while GPX4 0.05) downregulated. Oral squamous cell carcinoma (OSCC), one of the most prevalent and aggressive forms head neck carcinoma, has a five-year survival rate about 50% ~ 60%, emphasizing urgent need for more effective therapeutic strategies. Solute carrier family 7 member 11 (SLC7A11) is overexpressed in various cancers represents potential target. Sulfasalazine (SAS), Food Drug Administration-approved drug, potent inhibiter SLC7A11. However, SAS can also increase levels pro-inflammatory cytokines such as IL-1β, which may suppress immune response. Here, we investigate effect combined with anti-IL-1β monoclonal antibody (anti-IL-1β mAb) novel treatment strategy OSCC. In this study, SLC7A11 was markedly increased OSCC tissues, high expression predicted poor prognosis. shown to proliferation trigger ferroptosis, evidenced by elevated reactive oxygen species, reduced glutathione enhanced lipid peroxidation. IL-1β levels, leading T exhaustion. Combining mAb reversed exhaustion amplified anti-tumor effects vitro. 4-nitroquinoline-1-oxide-induced oral cancergenisis model, combination significantly inhibited carcinogenesis compared monotherapy. Our results suggest that combining enhances efficacy against through tumor growth inhibition modulation, offering promising strategy.
