ABSTRACT Ovarian endometrioma (OEM), a particularly severe form of endometriosis, is an oestrogen‐dependent condition often associated with pain and infertility. The mechanisms by which OEM impairs fertility, through its direct impact on oocyte‐cumulus cell (CC) communication related pathways, remain poorly understood. This study investigates the oocyte‐CC explores melatonin's therapeutic potential. We used mouse model employed ovarian transcriptome gene set enrichment analyses to identify disrupted alongside phalloidin staining for cytoskeletal analysis, gap junction coupling analysis intercellular communication, mitochondrial function assessments cellular metabolism. Our results showed that significantly steroidogenesis cumulus function, leading increased apoptosis, transzonal projections (TZPs), impaired antioxidant transfer oocytes. culminates in oxidative stress, dysfunction, compromised ATP production. oocytes also exhibited abnormalities, including DNA damage, maturation defects, spindle assembly disruptions, aneuploidy. identifies TZPs as key pathological feature highlights potential restore mitigate improve reproductive outcomes.

Oocyte maturation is a critical process responsible for supporting preimplantation embryo development and full to term. Understanding oocyte gene expression relevant given the unique molecular mechanism present in this gamete. Comparative transcriptome analysis across species offers powerful approach uncover conserved species-specific genes involved regulation of throughout evolution. Transcriptome identified 4,625, 3,824, 4,972 differentially expressed (DEGs) between germinal vesicle (GV) metaphase II (MII) stage human, porcine mouse oocytes respectively. These DEGs showed dynamic changes associated with maturation. Functional enrichment revealed that all three were mainly DNA replication, cell cycle redox regulation. 551 significant mitochondria mitochondrial intima. This study provides systematic comparative meiotic humans, pigs mice identifying both patterns during meiosis. Our findings also implied selection among these could form basis further exploring their functional roles human

ABSTRACT Gene edited pigs have extensive and important application value in the fields of agriculture biomedicine. With increasing demand medical research agricultural markets, more scenarios require gene to possess two or even advantageous phenotypes simultaneously. The current production multi is inefficient, time‐consuming, costly, there an urgent need develop efficient accurate editing technologies. polycistronic tRNA‐gRNA‐processing system (PTG), developed based on endogenous tRNA self‐processing systems, has been shown exhibit plants. This study aims combine a PTG strategy with multiple gRNA functions adenine base editor (ABE) test its feasibility for precise pig cells. results indicate that integrated ABE plasmid can perform at loci And while efficiency was significantly improved, no indel sgRNA dependent off target effects caused by DSB were detected. work permit will provide solid foundation applications.
