Metal-organic frameworks (MOFs) are widely used in the biomedical industry. The Western Ghats, India, is a hotspot for lichen diversity. However, the pharmacological importance of lichen-associated metabolites remains untapped. This study aimed to evaluate cytotoxic potential lichens this region. For this, sixteen macrolichens were collected and identified from two locations in Ghats. acetone extract Usnea cornuta (UC2A) showed significant cytotoxicity towards multiple human cancer cell lines. The oral cavity’s common pathologies are tooth decay, periodontal disease, and cancer; squamous cell carcinoma (OSCC) is the most frequent malignancy, with a high mortality rate. Our study aims to formulate, develop, characterize, pharmacologically investigate mucoadhesive patches (F-UBE-HPMC) loaded Usnea barbata (L.) F.H. Wigg dry ethanol extract (UBE), using HPMC K100 as film-forming polymer. Each patch contains 312 µg UBE, total phenolic content (TPC) of 178.849 33.924 usnic acid. Scanning electron microscopy (SEM) atomic force (AFM) were performed for their morphological characterization, followed by Fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD), thermogravimetric analysis (TGA). Pharmacotechnical evaluation involved measurement specific parameters follows: weight uniformity, thickness, folding endurance, tensile strength, elongation, moisture content, pH, disintegration time, swelling rate, ex vivo mucoadhesion time.
