Imidacloprid (IMI), a commonly utilized neonicotinoid insecticide, has been identified to adversely impact glucose homeostasis. Pregnant women are believed be more sensitive toxins than non-pregnant women, and the of IMI exposure on gestational hyperglycemia remain unclear. To explore impact, pregnant mice fed high-fat diet were exposed different doses (0.06, 0.6, 6 mg/kg bw/day) by gavage. Glucose homeostasis-related parameters measured. The homeostasis influenced treatment was explored through integrating gut microbiota, metabolomic transcriptomic analysis. Results showed that IMI-H (6 notably restricted weight gain perturbed characterized reduced tolerance insulin sensitivity, alongside elevated levels fasting blood insulin. Multi-omics analysis revealed induced significant changes in richness composition microbiome. metabolite profiles serum samples cecal contents, transcriptome liver ileum all affected treatment. altered metabolites genes exhibited correlations with parameters. These differential implicated various metabolic pathways including bile secretion, glucagon signaling pathway, lipid metabolism, fatty acid metabolism. Significant observed between microbiota caecum metabolome as well transcriptome. For example, abundance Oscillibacter strongly correlated microflora-related (Icosenoic acid, Lysosulfatide, fluticasone) (Grin3b, Lifr, Spta1). Together, resulted microbial composition, along alterations certain associated process, which may promote hyperglycemia.

This perspective presents first-hand research insights into the pervasive environmental distribution of neonicotinoid insecticides (NEOs) and their consequent human exposure. Our findings reveal widespread NEO detection across biological matrices, indicating potential health risks highlighting critical gaps in current risk assessment paradigms. We identify an urgent need for longitudinal epidemiological studies, particularly focusing on vulnerable populations, to comprehensively assess NEO-related outcomes. These scientific should inform evidence-based policy reforms management strategies effectively protect both public integrity.
