Abstract Ferroptosis, a unique modality of cell death with mechanistic and morphological differences from other modes, plays pivotal role in regulating tumorigenesis offers new opportunity for modulating anticancer drug resistance. Aberrant epigenetic modifications posttranslational (PTMs) promote resistance, cancer progression, metastasis. Accumulating studies indicate that can transcriptionally translationally determine vulnerability to ferroptosis functions as driver nervous system diseases (NSDs), cardiovascular (CVDs), liver diseases, lung kidney diseases. In this review, we first summarize the core molecular mechanisms ferroptosis. Then, roles processes, including histone PTMs, DNA methylation, noncoding RNA regulation such phosphorylation, ubiquitination, SUMOylation, acetylation, ADP-ribosylation, are concisely discussed. The PTMs genesis cancers, NSD, CVDs, well application PTM modulators therapy these then discussed detail. Elucidating mediated by will facilitate development promising combination therapeutic regimens containing or PTM-targeting agents inducers be used overcome chemotherapeutic resistance could prevent addition, highlight potential approaches chemoresistance halt

Neurodegenerative diseases such as Parkinson's disease (PD) have complex pathogenetic mechanisms. Genetic, age, and environmental factors are all related to PD. Due the unclear pathogenesis of PD lack effective cure methods, it is urgent find new targets for treating patients. Ferroptosis a form cell death that reliant on iron exhibits distinct morphological mechanistic characteristics compared other types death. It encompasses range biological processes, including iron/lipid metabolism oxidative stress. In recent years, research has found ferroptosis plays crucial role in pathophysiological processes neurodegenerative stroke. Therefore, also closely PD, This article reviews core mechanisms elucidates correlation between ferroptosis. addition, compounds emerged years exert anti effects by inhibiting signaling pathway were summarized. I hope further elaborate relationship through review this article, provide strategies developing treatments targeting

Abstract: Ferroptosis, a unique form of programmed cell death, is initiated by an excess iron accumulation and lipid peroxidation-induced damage. There growing body evidence indicating that ferroptosis plays critical role in the advancement tumors. The increased metabolic activity higher levels tumor cells make them particularly vulnerable to ferroptosis. As result, targeted induction becoming increasingly promising approach for cancer treatment. This review offers overview regulatory mechanisms ferroptosis, delves into mechanism action traditional small molecule inducers their effects on various In addition, latest progress inducing using new means such as proteolysis-targeting chimeras (PROTACs), photodynamic therapy (PDT), sonodynamic (SDT) nanomaterials summarized. Finally, this discusses challenges opportunities development ferroptosis-inducing agents, focusing discovering targets, improving selectivity, reducing toxic side effects. Ferroptosis is a type of iron-dependent programmed cell death characterized by depletion glutathione. Although generally less harmful to normal cells, in tumor the high demand for iron ions provides conditions conducive ferroptosis. In this review, we provide an overview recent progress research on regulation ferroptosis summarizing and assessing current state, trends, applications nanomaterials cells. Given advantages terms targeting, safety, improved drug efficacy, reduced side effects, these materials are considered have potential therapeutic value modulating cells via different mechanisms. respect, describe methods modifying interfering with glutathione activity lipid peroxidation. In the last decade, ferroptosis has received much attention from scientific research community. It differs other modes of cell death at morphological, biochemical, and genetic levels. Ferroptosis is mainly characterized by non-apoptotic iron-dependent caused lipid peroxide excess accompanied abnormal iron metabolism oxidative stress. recent years, more studies have shown that closely related to occurrence development lung diseases. COPD, asthma, injury, fibrosis, cancer, infection respiratory diseases become third most common chronic worldwide, bringing serious economic psychological burden people around world. However, exact mechanism which involved in progression not been fully revealed. this manuscript, we describe ferroptosis, targeting signaling pathways proteins, summarize relationship between diseases, explore intervention targeted therapy for

This study evaluated the effect of heat treatment on conversion ginsenoside and ameliorative heat-treated total (HG) from fresh ginseng cyclophosphamide (CTX)-induced liver injury. LC-MS analysis revealed that content rare ginsenosides increased markedly after treatment. HG significantly attenuated CTX-induced hepatic histopathological injury in mice. Western blotting showed untreated (UG) regulated Nrf2/HO-1 TLR4/MAPK pathways. Importantly, these results may be relevant to modulation intestinal flora. UG short-chain fatty acids (SCFAs)-producing bacteria Lactobacillus reduced LPS-producing Bacteroides Parabacteroides. These changes flora affected levels TNF-α, LPS SCFAs.
