High-efficient selection of aptamers by magnetic cross-linking precipitation and development of aptasensor for 1-aminohydantoin detection

Accurate
and
rapid
detection
of
nitrofurantoin
metabolite
1-aminohydantoin
(AHD),
as
one
the
tissue-bound
toxic
metabolites
in
foods,
is
crucial.
In
this
study,
a
fluorescent
aptasensor
was
constructed
for
AHD,
using
anti-AHD
aptamers
firstly
obtained
by
high-efficiency
magnetic
crosslinking
precipitation
(MCP)-SELEX.
The
AHD-specific
were
optimized
identified
their
binding
affinity
specificity
fluorescence-based
assays
molecular
dynamics
simulations.
truncated
aptamer
AHD50-1,
which
exhibited
highest
(Kd
=
86
nM)
an
excellent
specificity,
applied
to
construction
aptasensor.
Under
conditions,
showed
low
limit
(0.2
ng/mL)

dynamic
linear
range

ng/mL).
For
five
types
samples
(pork,
beef,
shrimp,
fish
honey),
recoveries
ranged
from
89.4%
110.04%,
with
coefficients
variation
ranging
1.43%
14.46%.
These
results
proposed
corroborate
very
well
method
HPLC
indicating
that
developed
provides
efficient
promising
sustainable
strategy
monitoring
AHD
animal
derived
food.
