As alternatives of perfluorooctanoic acid (PFOA), hexafluoropropylene oxide dimeric (HFPO-DA) and trimeric (HFPO-TA) have been detected increasingly in environmental media even humans. They shown to exhibit reproductive toxicity model species, but their effects on human remain unclear due the knowledge gap mode action. Herein, (anti-)androgenic two HFPOs PFOA were investigated underlying toxicological mechanism was explored by combining zebrafish test, cell assay molecular docking simulation. Exposure juvenile chemicals during sex differentiation promoted feminization, with HFPO-TA acting at an concentration 1 μg/L. The inhibited proliferation prostate cells transcriptional activity androgen receptors (AR), displaying strongest potency. Molecular revealed that bind AR a conformation similar known antagonist. Combined vivo, vitro silico results demonstrated disrupted likely antagonizing AR-mediated pathways, provided more evidence is not safe alternative PFOA.

With the prohibition of perfluorooctanoic acid (PFOA), emergence alternative perfluoroalkyl substances such as perfluorobutanonic (PFBA) and perfluorononanoic (PFNA) in various environmental matrices has led to concerns about their adverse effects on humans biota. This study aims investigate reproductive developmental toxicity PFBA PFNA by combined vivo, vitro, silico approaches. Examination juvenile zebrafish exposed at concentrations histopathology, sex hormone, gene expression revealed accelerated development toward males, while exposure during differentiation resulted feminization. In accordance with vivo results, activated androgen receptor (AR) signaling pathway, but inhibited it both prostate cancer cell proliferation luciferase reporter assays. Similarly, differential binding mode two chemicals AR was shown molecular docking analysis, exhibiting higher potency for agonist conformation favoring antagonistic conformation. Together, these results suggest that, exhibited similar activity PFOA, showed distinctive deserves particular attention further investigation.

The widespread contamination of perfluorooctanesulfonate (PFOS) has raised global concerns due to its adverse effects on human health. Consequently, there is an urgent need for sensitive, reliable, and cost-effective techniques the in-field detection PFOS in environmental matrices. Herein, we present a portable electrochemical sensor analysis, utilizing screen-printed electrode (SPE) modified with molecularly imprinted polymer (MIP). MIP layer specifically recognizes electrochemically inactive PFOS, hindering electron transfer between redox probe electrode, thereby providing sensitive robust strategy quantifying PFOS. Under optimized conditions, limit 9.25 ± 1.39 fM (4.98 × 10–3 ng/L) was achieved using potentiostat. Notably, interference from common coexisting ions analogs negligible. practical applicability demonstrated by measuring water samples, yielding results comparable those obtained established HPLC–MS/MS method while eliminating complex, time-consuming preconcentration procedures. MIP-modified SPEs holds significant potential as highly cost-effective, user-friendly technique monitoring, large-scale screening, early warning system water.

Ovarian cancer (OC) is the most lethal gynecological in developed world. Most cases are diagnosed at late stage III-IV with a very low 5-year overall survival rate. Several studies revealed an elevated risk of OC users hormone treatment (HT) compared non-users. The extended duration HT statistically significant factor. Carbohydrate antigen or 125 (CA-125) remains best screening tool for OC; however, its value limited due to specificity, leading unnecessary interventions, surgeries, and psychological harm. Additionally, variability ultrasound interpretation highlights urgent need develop univariate index higher sensitivity specificity early diagnosis women under HT. Herein we critically review limitations biomarkers detection aiming suggest accurate cost-effective diagnostic ratio that eliminates impact body mass index, age, HT, smoking, benign ovarian diseases on measurements.
