Introduction Although tripdiolide has demonstrated a protective role in lupus nephritis, its potential therapeutic and preventive effects on diabetic kidney injury remain inconclusive. Methods In this study, diabetes mice model was used to evaluate the effect of treatment kidney. The study assessed related factors levels, while comparing pathological changes, alterations intestinal microbiota composition, oxidative stress inflammation kidneys, validating cytokine expression protein pathway activation. Results experiment that effectively suppressed hyperglycemia elevated hemoglobin level, attenuated concentrations creatinine blood urea nitrogen, mitigated histopathological kidney, alleviated inflammatory cell infiltration. Tripdiolide regulated affected abundance Allobaculum , Dubosella Prevotella differential metabolic pathways primarily revolve around ubiquinol biosynthesis menaquinol biosynthesis. significantly renal mice, as evidenced by upregulation nuclear factor erythroid 2-related 2 (Nrf2), heme Oxygenase-1, downregulation phosphorylated factor-κB (P-NF-κB), NOD-like receptor 3. Experiments performed RAW264.7 cells tripdiolide. Discussion may play induced changing composition microorganisms, regulating Nrf2/NF-κB activation, inhibiting reaction. This contributes scientific evidence can inform development approaches for nephropathy.

In recent years, the study of gut microbiota has gradually become a research hotspot in field medicine, as dysbiosis is closely related to various diseases. Thalassemia, hereditary hemoglobinopathy, complex pathophysiological mechanism, and traditional treatment methods show limited efficacy. With deeper understanding microbiome, researchers have begun focus on its role pathogenesis thalassemia therapeutic effects. This article aims review potential prospects, analyze latest findings, explore impact mechanisms patients with thalassemia, goal providing new ideas directions for future clinical thalassemia.

Background Ulcerative colitis (UC) brings inconvenience to many patients with inflammatory bowel disease (IBD). Although colonic pathology is widely investigated, little attention has been paid the disorders in small intestine of UC. In this study, we investigated impairments UC and further explored how iron metabolism regulated epithelial integrity activity intestinal stem cells (ISCs). Methods Mice were treated by 2.5% dextran sulfate sodium (DSS) for 7 days established acute experimental colitis. Small tissues collected at different time points process DSS-induced Histological analysis was used evaluate changes intestine, including H&E, Alcian blue PAS staining, immunostaining, qRT-PCR. Iron content modulated supplementation ferric citrate or depletion deferoxamine (DFO). The influence on barrier cell function determined histology, IEC-6 cell, enteroid culture. ROS demonstrated DHE staining. proliferation (ISCs) shown BrdU Olfm4 Lgr5-tdTomato mice lineage tracing study. Results It that during colitis, underwent a serious injury process, dysregulated decreased ISCs. overload significantly exacerbated tissues, line, organoids. However, chelation (DFO) would greatly suppress injury. Mechanistically, generation enhanced infiltration immune cells. addition, STAT3 ERK pathways epithelium impaired interrupted expression p-STAT3 p-ERK1/2 within intestine. Conclusion summary, study proved also could affect damage regeneration, indicating strategy clinical practice needs be more cautious consider factors.

Background With the gradual rise of antibiotic-free farming practices, exploration novel, green, and low-pollution alternatives to antibiotics has become one key research focus in field agricultural science. In development antibiotic alternatives, probiotics, particularly host-associated have been found play a significant role enhancing production performance livestock poultry. However, on application probiotics specifically for meat pigeons remain relatively underdeveloped. Objective To assess investigate probiotic efficacy mechanisms during homologous lactic acid bacteria (LAB) transplant host-pigeons, LAB strains with good properties were isolated from intestinal contents 28-day-old Mimas pigeons. And then measured indexes, flora, transcriptomics hosts after instillation strains. Methods A total 360 at 1-day-old randomly divided into four groups gavaged 0.4 mL Ligilactobacillus salivarius S10 concentration 0, 10 8 , 9 CFU/mL, designated as control group (CG), low (LG), medium (MG), high (HG), respectively. Results The findings revealed that an optimal CFU/mL L. S10, dominant strain screened, enhanced growth young 16S rRNA gene sequencing analysis demonstrated increase abundance Lactobacillus, Pantoea_A Enterococcus_H reduction Clostridium_T pigeon ileum ( p < 0.05) under selected treatment. Transcriptomic profiling 1828 differentially expressed genes (DEGs) between CG MG. Notably, DEGs involved MAPK signaling pathway, such RAF1 PDGFRB ELK4 significantly correlated differential ileal bacteria, suggesting modulation flora can influence expression related cell proliferation differentiation ileum, which is potentially important promoting Conclusion possesses potential be used pigeons, gut development-related by regulating further improve This provides scientific foundation developing pigeon-specific promotes healthy practices Furthermore, it opens new avenues improving economic efficiency farming.
