Intestinal mucositis (IM) is a common adverse effect of chemotherapy, limiting its clinical application. Codonopsis pilosula-derived CP-A (an inulin-type fructan) an edible Chinese medicine with anti-inflammatory and gastrointestinal protective effects, which may be useful for treating IM. Here, we explored CP-A’s role in ameliorating IM induced by 5-fluorouracil (5-FU) investigated the underlying mechanism using vitro experiments rat models. Western blotting, immunohistochemistry (IHC), real-time PCR (RT-PCR) analyses were used to assess protein expression related extracellular-regulated kinases (ERK)/myosin light chain kinase (MLCK)/myosin 2 (MLC2) signaling pathway tight junction proteins. Inflammatory factors quantified enzyme-linked immunosorbent assays (ELISAs), 16S rRNA amplicon sequencing was employed cecum content analysis. The results indicated that restored body weight food intake reversed histopathological changes rats. Further, abnormal MLCK activation 5-FU attenuated via ERK/MLCK/MLC2 pathway. treatment improved levels reduced inflammatory factor expression. Moreover, intervention regulated intestinal microbiota community structure, increasing abundance Lactobacillus decreasing Shigella. In conclusion, mitigates 5-FU-induced inhibiting pathway, reducing factors, improving mucosal barrier, regulating microbial community. This study highlights therapeutic potential provides insights future research.

The spread of antibiotic resistance genes (ARGs) in agroecosystems through the application animal manure is a global threat to human and environmental health. However, adaptability colonization ability manure-derived bacteria determine pathways ARG agroecosystems, which have rarely been studied. Here, we performed an invasion experiment by creating synthetic communities (SynCom) with ten isolates from pig followed its assembly during gnotobiotic cultivation soil-Arabidopsis thaliana (A. thaliana) system. We found that Firmicutes SynCom were efficiently filtered out rhizosphere, thereby limiting entry tetracycline (TRGs) into plant. Proteobacteria Actinobacteria able establish all compartments soil–plant system spreading TRGs soil presence native prevented establishment manure-borne effectively reduced TRGs. Achromobacter mucicolens Pantoea septica main vectors for tetA plants. Furthermore, doxycycline stress promoted horizontal gene transfer (HGT) conjugative plasmid RP4 within A. upregulating expression HGT-related mRNAs. Therefore, this study provides evidence dissemination ARGs agricultural systems HGT plasmids demonstrates priority rhizosphere limited soil-plant systems.

The impact of antibiotic therapy on the spread resistance genes (ARGs) and its relationship to gut microbiota remains unclear. This study investigated changes in ARGs, mobile genetic elements (MGEs), microbial composition following tilmicosin administration pigs. Thirty pigs were randomly divided into control (CK), low-concentration (0.2 g/kg; L), high-concentration (0.4 H) groups. Tilmicosin concentration manure peaked day 16 dosing dropped below detectable levels by 13 withdrawal period. While did not significantly affect total abundance macrolide (MRGs) (p > 0.05), it increased multidrug gene tolC H group compared with L CK groups during period < 0.05). increase was associated a coincidental rise MGEs (e.g., int1 int2) growth potential tolC-hosting bacteria such as Paenalcaligenes Proteiniclasticum. Redundancy analysis showed primary driver MRG abundance, MGEs, concentration, physicochemical properties playing secondary roles. These findings suggest that high-dose may alter promote ARG via MGE-mediated transfer.
