Acinetobacter baumannii and Klebsiella pneumoniae are significant nosocomial pathogens worldwide. In this study, the general characterization of A. K. isolates obtained from blood intensive care unit patients multidisciplinary scientific practical center emergency medicine January to September 2024 was performed. Prophage regions prophage-derived tailspike polysaccharide-depolymerizing or -modifying enzymes within these were identified characterized in detail using a refined workflow. The protocol, encompassing comprehensive survey all predicted bacterial proteins, revealed an average 6.0 prophage per genome, including putatively derived filamentous phages, 4.8 isolate. Acinetobacter baumannii, a notorious opportunistic pathogen, presents formidable challenge in both clinical and environmental fields due to its resilience ability acquire resistance. This study undertook comprehensive analysis of 183 A. baumannii isolates collected between 2019 2022 from intra-hospital infections (IHI), hospital sewages (Hs), wastewater treatment plants (WWTP), adjacent river waters two Southern cities, focusing on their resistome, virulome, mobilome through isolation chromogenic media, identification by MALDI-TOF-MS antibiotic susceptibility testing disk diffusion) followed genotypic characterization [whole-genome sequencing (WGS), 3rd generation the MinION (ONT) platform, pangenome description, respectively horizontal gene transfer conjugation assays]. Introduction The extent to which sinks are contaminated by carbapenem-resistant Acinetobacter baumannii (CRAB) in intensive care units (ICUs) and the association between these hospital-acquired CRAB infections during non-cluster period remains largely unknown. Here, we performed a prospective multicenter study 16 ICUs at 11 tertiary hospitals Chengdu, China. Methods We sampled sinks, collected clinical isolates, conducted whole-genome sequencing analysis. Results A total of 789 swabs were from 158 isolates recovered resulting contamination rate 10.16%. Twenty-seven period. majority (97.67%, 42/43) belonged ST2, 36 (83.72%) them had both bla OXA-23 OXA-66 . 43 strains 12 clones. One certain clone caused multiple contaminations seven one GICU. Two clones ST2 -carrying sink likely sources two clusters GICUs, respectively. Five found be common but hospitals. Conclusion handwashing is high some local ICUs, can serve as environmental reservoirs for clusters.

ABSTRACT Acinetobacter, particularly the Acinetobacter baumannii group, is a major cause of nosocomial infections, and carbapenem-resistant spp. are important human pathogens. We collected 492 strains from two hospitals in Taiwan classified them using MALDI-TOF MS bla OXA-51-like PCR; 94.5% were A. baumannii, 5.5% non- A . (NAB). confirmed their identity by rpo B gene sequencing 239 randomly selected all 27 NAB strains. Our analysis revealed that alleles OXA51-like-negative matched those species, seifertii nosocomialis , while OXA51-like-positive as per Pasteur MLST scheme database. Among strains, 240 exhibited carbapenem resistance, including 237 (CRAB) three CR-NAB All CRAB positive for ; 72.6% also carried OXA-23-like 22.8% OXA-24-like 3.4% co-carried + 1.27% alone. one NDM-1 OXA-58-like IMP established new multiplex PCR method rapid screening common capsular types (KL), which showed difference between carbapenem-susceptible (CSAB).
