The effects of perfluoroalkyl and polyfluoroalkyl substances (PFAS) on genomic stability remain unclear. Here, a cross-sectional study was conducted to establish the associations PFAS with genotoxic biomarkers. We recruited cohort 453 residents in 2021 Zhejiang, China. Thirty serum were quantified, alongside seven indicators [five rDNA copy numbers (rDNA-CN), mitochondrial DNA (mtDNA-CN), relative telomere length (RTL)] whole blood. Results showed that PFUnDA, perfluorohexanesulfonic acid (PFHxS), perfluorooctanesulfonic (PFOS), 6:2 Cl-PFESA, PFO5DoDA positively correlated rDNA-CN, while PFHpA, PFOA, PFMOAA inverse associations. PFO4DA mtDNA-CN. HFPO-TA, negatively associated RTL, perfluorononanoic acid, PFHxS, PFOS, Cl-PFESA positive Nonlinear exposure-response relationships also observed between biomarkers using restricted cubic spline models. Furthermore, mixtures mtDNA-CN, showing highest contribution by quantile-based g-computation model. In vivo studies further confirmed increased mtDNA-CN male mice dose-dependent manner. This provides novel evidence disrupt stability, varying functional groups fluoroalkyl(ether) chain lengths.
