Abstract The incidence of thrombosis-related diseases is increasing globally, and it often treated by mechanical thrombectomy, intravenous thrombolysis antiplatelet, but easy to lead drug resistance bleeding complications, which limits the development its treatment. gut-liver axis represents a complex two-way communication system, in intestinal microorganisms not only affect immunity function, also regulate coagulation function liver. Jingfang Mixture (JF) Traditional Chinese Medicine (TCM) that have antithrombotic, mechanism still unclear. This study aimed explore regulatory effect JF on thrombosis rats through carrageenan-induced rat model. results showed after treatment, degree black tail with was significantly reduced, pathological condition improved, four parameters levels factors were adjusted closely normal levels, liver inflammatory reduced. In vitro experiments proved had little platelet aggregation, release, coagulation. our study, can elevate tight junction proteins improve flora disorders, alleviate oxidative stress level liver, short-chain fatty acid feces thrombosis. Of note, relative abundances Clostridia_UCG-014, Ruminococcaceae, Eggerthellaceae, Monoglobaceae Rikenellaceae decreased JFM intervention. Proteomic antithrombotic may be related complement cascade signaling pathway. gene protein expression FGA, FGB, FGG C3 elevated respectively. Finally, we further confirmed therapeutic fecal microbiota transplantation (FMT) experiments, crosstalk contributed improvement summary, this took as breakthrough point coupled 16S rDNA sequencing, non-targeted metabolomics, TMT-based quantitative proteomics comprehensive analysis methods, showing immune system disorder, well reduce damage rats, protective accurately explains multi-target multi-pathway treating diseases, provide reference for clinical application.
