In recent years, deep eutectic solvents (DESs) have garnered significant attention as promising green alternatives to conventional organic for a wide range of applications. this study, four novel polyethylene glycol (PEG)-based DESs were prepared and evaluated their physicochemical properties, including density, dynamic viscosity, kinematic viscosity. Fourier transform infrared spectroscopy (FT-IR) NMR analyses revealed substantial intermolecular interactions between the hydrogen bond donor acceptor components, confirming formation stable DES systems. The application was tested in biological separation, specifically selective extraction bovine serum hemoglobin (BHb). This study demonstrates efficacy PEG-based selectively extracting BHb. Among studied, DES-4 (PEG-600) achieved highest efficiency 88%, while maintaining protein stability. Spectroscopic techniques, UV–visible, fluorescence, light scattering, circular dichroism, FT-IR, employed investigate mechanism, conformational changes structure, DES-protein interactions. These methods provided insights into structural stability functionality BHb during process. characterizations confirmed unique properties DESs, making them viable candidates sustainable extraction. Their compatibility, excellent efficiency, short separation times underscore potential environmentally friendly long-lasting substitutes techniques. highlights advancement chemistry biotechnological applications, offering an efficient platform integrity.
