Decades of research have investigated the mechanisms that lead to origin cancer, striving identify tumor-initiating cells. These cells, also known as cancer stem are characterized by ability self-renew, give rise differentiated tumor populations, and on a larger scale, deemed responsible not only for initiation but recurrent tumors, often resistant chemotherapy radiotherapy. Long noncoding RNAs RNA molecules longer than 200 nt, lacking code proteins, with recognized roles fine regulators gene expression. They can exert these functions through variety mechanisms, acting at almost all steps expression, from modulation epigenetic state chromatin protein stability. In cases, lncRNAs do work alone, they always interact other molecules, either coding or non-coding, factors. this review, we summarize latest results obtained about involvement in initiating cells several types highlight different which work, while discussing how lncRNA affect aspects onset progression.

Genetic studies implicate the oncogenic transcription factor Forkhead Box M1 (FOXM1) as a potential therapeutic target in high-grade serous ovarian cancer (HGSOC). We evaluated activity of different FOXM1 inhibitors HGSOC cell models. treated and fallopian tube epithelial (FTE) cells with panel previously reported inhibitors. Based on drug potency, efficacy, selectivity, determined through viability assays, we focused two compounds, NB-73 NB-115 (NB compounds), for further investigation. NB compounds potently selectively inhibited lesser effects other FOX family members. decreased expression via targeting protein by promoting its proteasome-mediated degradation, effectively suppressed gene targets at both mRNA level. At cellular level, promoted apoptotic death. Importantly, while inhibition apoptosis using pan-caspase inhibitor rescued from compound-induced death, it did not rescue supporting that loss compound treatment is specific general consequence cytotoxicity. Drug washout indicated reduction was retained least 72 h post-treatment, suggesting exhibit long-lasting cells. two-dimensional three-dimensional colony formation sub-micromolar concentrations. Abstract The proliferation-specific oncogenic transcription factor, FOXM1 is overexpressed in primary and recurrent breast tumors across all cancer (BC) subtypes. Intriguingly, overexpression was found to be highest Triple-negative (TNBC), the most aggressive BC with worst prognosis. However, FOXM1-mediated TNBC pathogenesis not completely elucidated. Single nucleotide polymorphisms (SNPs) are common genetic variations causing functional structural aberrations proteins enhancing susceptibility. This computational investigation attempted identify malignant non-synonymous SNPs (nsSNPs) evaluate their role affecting conformational stability, evolutionary conservation, post-translational modifications, susceptibility of protein. Out a huge data pool 8826 using several in-silico sequence-based tools approaches, four viz. E235Q, R256C, G429E S756P were identified as pathogenic nsSNPs among shortlisted variants molecular dynamics simulations E235Q damaging SNP, followed by S756P. Additionally, defective drug DNA binding motif also determined our study. Thus, although further in-vitro validations awaited findings this work can used blueprint for nsSNP identification aiding clinical therapeutics.
