Background/Objectives: Monoclonal gammopathies, such as Gammopathy of Undetermined Significance (MGUS) and Smoldering Multiple Myeloma (SMM), are conditions marked by the overproduction specific monoclonal proteins. Patients with these known to have a higher risk fractures compared general population, yet there no established guidelines for assessing or managing their skeletal health. The Trabecular Bone Score (TBS), which can be calculated from DXA images lumbar spine, provides additional insights into bone microarchitecture. Methods: This study aimed determine whether TBS serve supplementary tool in loss MGUS SMM patients. Conducted 2020 2023, involved 148 participants—74 diagnosed myeloma precursor state 74 healthy controls—who underwent simultaneous measurements. Results: results indicated weak positive correlation (R = 0.405) between T-scores, suggesting that other factors may influence When analyzed separately, correlations remained both 0.250) 0.485). Interestingly, discrepancies were noted T-score classifications; instance, patient classified normal via could deemed osteopenic osteoporotic TBS. Conclusions: Overall, findings suggest incorporating alongside enhance accuracy density assessments, facilitating earlier diagnosis treatment initiation osteoporosis asymptomatic patients gammopathies.

Purpose Based on the efficacy data from previous study of B-cell maturation antigen (BCMA) chimeric receptor (CAR) T-cell injection, we further examined delayed toxicity for 8 weeks after a single dose BCMA CAR injection to observe possible toxic reactions. Methods B-NDG mice transplanted with multiple myeloma (MM) cells were given at two dosages or human normal T and then subjected examinations including clinical signs, weight food intake measurements, haematology, blood biochemical analysis, cytokine assay, T-lymphocyte subpopulation quantification histopathology days 28 56 dosing. In addition, quantitative polymerase chain reaction (qPCR) was used quantify DNA fragments in different tissues assess tissue distribution provide basis its preclinical safety evaluation Results study, no mortality significant effects such as reductions intake, body weight, relevant parameters target organ weights observed T-cell-treated groups. Compared model group, restorative changes signs clinicopathology indicating therapeutic seen Human-derived cytokines interleukin-2 (IL-2), IL-4, IL-6, IL-12, IL-10, tumor necrosis factor α (TNF-α), interferon-γ (IFN-γ) could be detected all cancer cell–bearing by level measurement. IFN-γ levels showed geometric increase due graft versus host disease (GVHD) response induced mice, while other did not show changes. Histopathological examination indicated that treatment groups mixed cellular infiltration human-derived cells, inflammatory several organs liver, spleen, lung, kidney, some them mild damage, but number animals severity damage significantly less than those control group well group. The results mainly concentrated bone marrow related immune organs/tissues, highly consistent MM suggesting follow during metastatic targeting tissues. Conclusions present demonstrated low manageable side good anticancer activity without observable adverse effects. This provides support future studies MM.

To evaluate prognostic factors in women with invasive VSCC at Sun Yat-sen University Cancer Center (SYSUCC). 137 patients SYSUCC were retrospectively analyzed. The Kaplan-Meier method assessed the overall survival (OS) and progression-free (PFS) time. Prognostic identified using univariable multivariable Cox regression analysis. Only 2 out of had positive postoperative margins after intraoperative supplemental excision. international federation gynecology obstetrics (FIGO) Stage III-IV (HR: 4.67, 95 % confidence intervals (CI): 2.48-8.79) BMI ≥25 kg/m2 1.86, CI: 1.08-3.23) independent risk for OS. affecting PFS included FIGO stage 3.72, 2.10-6.60), 2.15, 1.28-3.64), squamous cell carcinoma antigen (SCC-Ag) > 1.5 ng/ml 2.06, 1.23-3.47). 12 individuals perineural invasion (PNI) was extremely poor, a median OS 37 months 22 months. surgical margin should be least 1.0 cm away from tumor edge. When surgeons cannot ensure negative margins, detecting rapid pathological examination may reduce incidence margins. III-IV, ILN metastases, are important adverse patients. Cases PNI have poor prognosis. SCC-Ag might useful marker predicting relapse.
