Aminopeptidases, enzymes with critical roles in human body, are emerging as vital biomarkers for metabolic processes and diseases. Aberrant aminopeptidase levels often associated diseases, particularly cancer. Small-molecule probes, such fluorescent, fluorescent/photoacoustics, bioluminescent, chemiluminescent essential tools the study of aminopeptidases-related The fluorescent probes provide real-time insights into protein activities, offering high sensitivity specific locations, precise spatiotemporal results. Additionally, photoacoustic offer signals that able to penetrate deeper tissues. Bioluminescent can enhance vivo imaging abilities by reducing background. This comprehensive review is focused on small-molecule respond four key aminopeptidases: N, leucine aminopeptidase, Pyroglutamate 1, Prolyl Aminopeptidase, their utilization tumors afflicted regions. In this review, design strategy variety designs from previous studies, opportunities future bioimaging applications discussed, serving a roadmap research, sparking innovations aminopeptidase-responsive probe development, enhancing our understanding these disease diagnostics treatment.
