Assessing the activity of DNA methyltransferases (MTases) and screening for methyltransferase inhibitors not only allow a deep exploration role methylation regulation in disease initiation progression but also provide an important experimental clinical basis diagnosis treatment diseases. Herein, new COFs functionalized electrochemical biosensor has been developed to detect adenine (Dam) MTase with high sensitivity rapidity by taking advantage walker rolled circular strand displacement amplification (RC-SDA) reaction. Specifically, hairpin probe H1 was methylated Dam MTase, followed site-specific cleavage DpnI enzyme generate S5 probe. The padlock probes two nucleic acid endonuclease sites were introduced trigger RC-SDA reaction under action primer, releasing large number single-stranded S6 probes. released worked synergistically Exo III walking, which exposed binding enable Au-COF-MB signal bind effectively electrode surface, signals thus generated. result showed that designed demonstrated excellent specificity detecting detection limit (LOD) 6.85 × 10-5 U/mL. This method provides support development strategies related
