Clear cell renal carcinoma (ccRCC) is a prevalent urogenital malignancy characterized by heterogeneous patterns. Stemness pivotal factor in tumor progression, recurrence, and metastasis. Nevertheless, the impact of stemness-related long non-coding RNAs (SRlncRNAs) on prognosis ccRCC remains elusive. In this study, we aimed to delve into SRlncRNAs develop signature for risk stratification prediction. Gene-expression clinical data were downloaded from The Cancer Genome Atlas (TCGA) Gene Expression Omnibus (GEO) databases. We calculated RNA stemness scores (RNAss) samples evaluate their stemness. mRNAs (SRmRNAs) identified through weighted correlation network analysis (WGCNA), which employed sophisticated statistical methodologies identify interconnected modules related genes. ABSTRACT Digestive system tumor, including esophageal gastric intestinal liver pancreatic and cholangiocarcinoma, are the most common tumors worldwide serve as a major cause of tumor‐related death. Cancer stem cells (CSCs) small group in that harbor self‐renewal, differentiation abilities, playing crucial role tumor initiation, progression, metastasis, supposed to be fundamental recurrence after conventional treatment. A comprehensive understanding targeting CSCs is key overcoming tumors. In this review, focusing on digestive tumors, we summarize characteristics CSCs, review intracellular mechanisms regulate self‐renewal functional maintenance stemness pathways, transcription epigenetic regulation, metabolic noncoding RNAs, demonstrate microenvironmental regulation systemic at molecular cellular levels. Abstract Tumor immunotherapy has significantly transformed the field of oncology over past decade. An optimal tumor would ideally elicit robust innate and adaptive immune responses within microenvironment (TIME). Unfortunately, system experiences functional decline with chronological age, a process termed “immunosenescence,” which contributes to impaired against pathogens, suboptimal vaccination outcomes, heightened vulnerability various diseases, including cancer. In this context, we will elucidate hallmarks molecular mechanisms underlying immunosenescence, detailing alterations in immunosenescence at molecular, cellular, organ, disease levels. The role tumorigenesis senescence‐related extracellular matrix (ECM) also been addressed. Recognizing that is dynamic influenced by factors, evaluate treatment strategies targeting mechanisms, as well methods for cell, organ restoration, present emerging approaches immunotherapy. overarching goal research prevent development, recurrence, metastasis, ultimately improving patient prognosis.
