Pandan, a tropical crop, is rich in squalene (SQ), known for its antioxidant and hypoglycemic properties, 2-acetyl-1-pyrroline (2-AP), which imparts characteristic aroma. This study focuses on the extraction of two bioactive compounds from Pandan leaves investigates effects drying methods, solvents, conditions yield SQ 2-AP. Results show that hot air-dried when extracted using binary solvent system ethanol n-hexane (EH), higher content while maintaining an adequate To further optimize process, single-factor experiment was followed by optimization Box-Behnken design (BBD) response surface methodology (RSM). The optimal were determined as follows: ultrasound time 60 min, temperature 50 °C, power 300 W, solid-to-liquid ratio 1:5 g/mL. Under these conditions, 1229.98 ± 13.09 μg/DW 1 g 2-AP 80.72 0.88 achieved, representing increases 3.30% 9.82% compared to pre-optimization values. Additionally, activities EH extracts evaluated through various vitro assays. demonstrated significant DPPH ABTS free radical scavenging activity (12.46 μmol TE/g DW 22.14 DW, respectively), along with ferric cupric ion reducing (10.629 14.275 respectively). also exhibited notable inhibitory α-amylase α-glucosidase. findings suggest are promising natural source antioxidants potential applications health nutrition.
