Abstract The intricate morphology, physicochemical properties, and interacting proteins of lipid droplets (LDs) are associated with cell metabolism related diseases. To uncover these layers information, a solvatochromic photosensitized LDs‐targeted probe based on the furan‐based D‐D‐π‐A scaffold is developed to offer following integrated functions. First, turn‐on fluorescence upon selectively binding LDs allows for direct visualization their location morphology. Second, its linear correlation polarity quantifies micro‐environmental heterogeneity among LDs. Third, unique properties enable photocatalytic proximity labeling enrichment LDs‐interacting proteins, ready potential downstream proteomic analysis. These functions exemplified using artificial in buffer, stressed liver line, diseased tissues biopsied from patients. While most LD sensors only imaging functions, multi‐functional reported herein integrates both singlet triplet photosensitization studies.
