This study aims to evaluate the effects of melatonin on primordial follicle activation and survival, stromal cell density collagen distribution in extracellular matrix (ECM). The involvement receptors mTORC1 pathway these processes were also investigated. To this end, ovarian fragments culture for six days α‐MEM + alone or supplemented with 1000 pM melatonin, both luzindole (inhibitor receptors), 0.16 µg/ml rapamycin (mTORC1 inhibitor). At end period, tissues processed classical histology follicles classified as normal degenerated, well growing follicles. ECM evaluated. Samples destined measure levels thiol mRNA CAT, SOD, GPX1 PRDX1, activity antioxidant enzymes GPX1. results showed that cultured had significantly higher percentage morphologically than those control medium (α‐MEM+). presence either rapamycin, however, did not block positive survival (P>0.05). reduced increased development follicles, but blocked by (P<0.05). Melatonin, influence number cells. In contrast, percentages tissues, rapamycin. Tissues CAT lower GPx when compared medium. conclusion, promotes activation, increases fiber vitro bovine tissue through its membrane-coupled mTORC1. On other hand, increase acting pathways, since it can pass membranes directly regulate oxidative stress.

Non-communicable diseases (NCDs) and premature aging, caused by free radicals, have spurred a demand for extensive research into finding effective antioxidants. Currently, there is an abundance of both natural synthetic antioxidants, including metal nanoparticles with high antioxidant activity. Among these, gold (AuNPs) stand out as favoured antioxidants because their minimal toxicity, simple synthesis, detectability. The properties AuNPs enhance its wide-ranging potential use in healthcare applications anti-aging, anti-inflammatory, wound healing agents, well treatment various diseases. This review highlights recent progress the synthesis method assessing capacity delves mechanism action explores health applications. Oxidative stress during in vitro of ovarian tissues has adverse effects on follicle survival. α-pinene is a monoterpenoid molecule with antioxidant activity that great potential to maintain cell survival vitro. This study investigated the effect (1.25, 2.5, 5.0, 10.0, or 20.0 μg/mL) primordial growth and morphology, as well stromal cells collagen fibers bovine slices cultured for six days. The transcripts catalase (CAT), superoxide dismutase (SOD), peroxiredoxin 6 (PRDX6), glutathione peroxidase (GPX1), nuclear factor erythroid 2-related 2 (NRF2) was by real-time PCR. were processed histological analysis evaluate follicular growth, density, fibers. results showed 10.0 µg/mL increased percentages normal follicles but did not influence growth. (10.0 µg/mL) kept density levels tissue like uncultured tissues. Ovarian control medium had reduced expression mRNA NRF2, SOD, CAT, GPX1, PRDX6, NRF2 PRDX6. In conclusion, improves survival, preserves levels, increases PRDX6 after culture tissue.

This study aimed to investigate the changes in extracellular matrix and number of stromal cells around primordial primary follicles culture bovine ovarian tissues. Furthermore, follicular oocyte diameters, granulosa cell survival, activity antioxidant enzymes were assessed. To this end, fragments cultured α-minimum essential medium (α-MEM+) for 6 days. Before (uncultured control) after culturing, fixed, processed histological analysis stained with hematoxylin eosin assess survival measure diameters. analyze collagen fibers glycosaminoglycans, sections picrosirius red or Alcian blue. Uncultured tissue samples also used mRNA expression superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPX), peroxiredoxin (PRDX). The thiol levels CAT, SOD, GPX investigated. results demonstrated that, 6-day tissues, GAGs network exhibited a great degree disorganization, deposition these components surrounding was significantly reduced when compared uncultured (P<0.0001). (P<0.05) (P<0.0001) In addition, number, as well diameters both categories Regarding protection, GPX, PRDX downregulated Similarly, CAT reduced. conclusion, vitro affects abundance organization glycosaminoglycans network, reduces stroma follicles. resulted lower proportion health cells, protection.
