Malvidin (MV), an essential anthocyanin, has antioxidant and anti-inflammatory effects that may help treat pulmonary fibrosis (PF), a progressive occasionally fatal condition characterized by severe lung scarring, oxidative stress, inflammation. This study aims to evaluate the therapeutic potential of MV in PF assessing its on inflammation, fibrotic markers through vitro vivo models. The compound was evaluated molecular docking. BEAS-2B RLE-6TN cells were treated with 200 µg/mL BLM induce followed treatment. Cell viability, ROS levels, wound healing analyzed. Abstract This study is aimed at investigating the possible protective effect of diacerein (DIA) against AMD-induced pulmonary fibrosis in rats. Rats were classified into 4 groups: a normal group that received distilled water, control AMD (100 mg/kg, p.o.) for 21 days to induce fibrosis, and 2 treatment groups diacerein, dose levels (50 100 p.o., respectively) addition p.o.), days. Lung function test was assessed using spirometer; serum tissue collected. Biochemical, real-time PCR, histopathological, immunohistopathological analyses carried out. reduced tidal volume (TV), peripheral expiratory rate (PER), forced vital capacity (FVC), glutathione (GSH) levels, Beclin, LCII, while it elevated transform growth factor (TGF-β1) gene expression, malondialdehyde (MDA) level, alpha-smooth muscle actin (α-SMA), Smad3, phosphorylated signal transducer activator transcription (p-STAT3), p62 lung content. Also, tumor necrosis factor-alpha (TNF-α) caspase-3 protein expression. DIA TV, PER, FVC, GSH TGF-β1 MDA α-SMA, p-STAT-3, Moreover, TNF-α attenuated via alleviating TGF1/α-SMA/Smad3 pathway, reducing STAT-3 activation, combating oxidative stress inflammation promoting autophagy abrogating apoptosis.
