Efficient and sensitive detection of targets is one the motivations for constant development innovation various biosensors. CRISPR-Cas12a, a new generation gene editing tools, has shown excellent application potential in biosensor design construction. By combining with specific recognition element-aptamer, single-stranded oligonucleotide obtained by systematic evolution ligands exponential enrichment (SELEX) vitro screening, CRISPR-Cas12a also shows superior performance non-nucleic acid detection, such as small molecules, proteins, virus pathogenic bacteria. However, aptamer (CRISPR-Cas12a/Apt) still face some problems target single signal response mode narrow linear range. The diverse CRISPR-Cas12a/Apt biosensors necessary to meet needs environments. In this review, working principle was introduced recent progress summarized. Moreover, requirements critical parameters crRNA sequence, activator reaction system were discussed, which could provide reference efficient novel addition, challenges prospects CRISPR-Cas12a/Apt-based further presented.

Food safety is a global issue in public hygiene. The accurate, sensitive, and on-site detection of various food contaminants performs significant implications. However, traditional methods suffer from the time-consuming professional operation, restricting their application. Hydrogels with merits highly porous structure, high biocompatibility, good shape-adaptability, stimuli-responsiveness offer promising biomaterial to design sensors for ensuring safety. This review describes emerging applications hydrogel-based inspection recent years. In particular, this study elaborates on fabrication strategies unique sensing mechanisms depending whether hydrogel stimuli-responsive or not. Stimuli-responsive hydrogels can be integrated functional ligands sensitive convenient via signal amplification transduction; while non-stimuli-responsive are mainly used as solid-state encapsulating carriers probe, nanomaterial, cell conductive media. addition, existing challenges, future perspectives, application prospects discussed. These practices greatly enrich scenarios improve performance detection.
