Abstract In this narrative review, we assess the pathophysiology of severe adverse events that presented after vaccination with DNA and mRNA vaccines against COVID‐19. The focus is on perspective an undersulfated degraded glycocalyx, considering its impact immunomodulation, inflammatory responses, coagulation oxidative stress. paper explores various factors lead to glutathione inorganic sulfate depletion their subsequent effect glycocalyx sulfation other metabolites, including hormones. Components COVID‐19 vaccines, such as material, spike protein antigen lipid nanoparticles, are involved in possible cytotoxic effects. common thread connecting these endotheliopathy or degradation, caused by depleted levels, shear stress from circulating aggregation formation coronas; leading imbalanced immune responses chronic release pro‐inflammatory cytokines, ultimately resulting systemic response syndrome. By understanding underlying events, better treatment options can be explored.

Abstract Clinical data indicate that COVID-19 causes cardiovascular complications, regardless of the severity disease. In this work, we have shown SARS-CoV-2 infection vascular dysfunction due to modification endothelial cell elasticity. We used human pulmonary cells (HPAECs) expressing ACE2 receptor as a model endothelium. This system mimics in vivo conditions, it allows virus entry but not replication. As reference, A549 epithelial cells, well-described supports productive replication SARS-CoV-2. show HPAECs results loss elasticity, which correlates with increased polymerization actin filaments and induction inflammatory response. On contrary, supporting viral showed also elasticity were impaired after Alpha, Beta Delta variants. Consequently, believe nonproductive associated endothelium may be clinically relevant result damage tissue.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection has been associated with systemic inflammation and vascular injury, which contribute to the development of (ARDS) mortality COVID-19 infection. Moreover, multiorgan complications due persistent endothelial dysfunction have suspected as cause post-acute sequelae SARS-CoV-2 Therefore, elucidation inflammatory effect will increase our understanding how cells (ECs) short- long-term consequences Here, we investigated interaction spike protein human ECs from aortic (HAoEC) pulmonary microvascular (HPMC) origins, cultured under physiological flow conditions. We showed that triggers prolonged expression cell adhesion markers in both ECs, similar TNF-α. treatment also led release various cytokines chemokines observed severe patients. increased binding leucocytes surface a procoagulant state endothelium were observed. Transcriptomic profiles spike-activated HPMC HAoEC upregulation genes pathways responses virus, cytokine-mediated signaling, pattern recognition, well complement coagulation pathways. Our findings support experimental clinical observations highlight importance EC protection one strategies mitigate effects possible disease.
