Abstract Decades of extensive research have documented the presence neural innervations sensory, sympathetic, or parasympathetic origin in primary and secondary lymphoid organs. Such inputs can release neurotransmitters neuropeptides to directly modulate functions various immune cells, which represents one essential aspects body's neuroimmune network. Notably, recent studies empowered by state‐of‐the‐art imaging techniques comprehensively assessed distribution patterns BM, thymus, spleen, LNs rodents humans, helping clarify several controversies lingering field. In addition, it has become evident that organs are not static but undergo alterations pathophysiological contexts. This review aims update current information on neuroanatomy obtained through whole‐tissue 3D genetic approaches, focusing anatomical features may designate functional modulation responses. Moreover, we discuss critical questions call for future research, will advance our in‐depth understanding importance complexity control

Communication between the nervous system and immune has evolved to optimally respond potentially dangerous stimuli both from within outside body. Tumors pose a severe threat an organism current therapies are insufficient for tumor regression in majority of cases. Studies show that tumors innervated by peripheral nerves sensory, parasympathetic sympathetic systems. Interactions cancer cells, cells regulate overall progression. Clinical studies have indicated potential targeting promoting anti‐tumor responses. This view point provides opinion on evidence therapeutic manipulating neuro‐immune communications cancer.

Sensory neurons cooperate with barrier tissues and resident immune cells to form a significant aspect of defensive strategies in concert the system. This assembly neuroimmune cellular units is exemplified across evolution from early metazoans mammalian life. As such, sensory possess capability detect pathogenic infiltrates at surfaces. capacity relies on mechanisms that unleash specific cell signaling, trafficking reflexes. These pathways exploit amplify enhance alerting response should infiltration seep into other tissue compartments and/or systemic circulation. Here we explore two hypotheses: 1) neurons’ potential signaling require interaction pathogen recognition receptors ion channels and; 2) which these sensing activation multiple neuron sites. Where possible, provide references apt reviews reader more detail aspects perspectives provided here.
