Cadmium (Cd) is a non-essential heavy metal that highly toxic to testicle. Selenium (Se) known possess antagonistic effects against Cd toxicity, yet the precise mechanisms through which Se counteracts Cd-induced testicular damage in chickens Nuclear factor erythroid 2-related 2/Heme oxygenase-1 (Nrf2/HO-1) signaling pathway, oxidative stress (OS), apoptosis, and inflammation remained unclear. In present study, experimental model of chicken testis was established by incorporating CdCl2 Na2SeO3 into dietary intake. After 60 days, from each group were euthanized, serum samples subsequently collected. Ultrastructural assessment revealed supplementation significantly mitigated induced Cd. effectively suppressed elevation ROS, MDA, H2O2 levels, while also preventing downregulation CAT, GSH, T-AOC levels. Furthermore, administration ameliorated reduction expression levels Nrf2, HO-1, Bcl-2 Cd, counteracted overexpression Caspase-3, Bax, Cyt-c, Caspase-9, TNF-α, IL-2, IL-6, IL-1β. Meanwhile, immunofluorescence data demonstrated attenuated decrease Nrf2 HO-1 upregulation IL-6 expression. conclusion, this study elucidated might mitigate testicles stimulation Nrf2/HO-1 thereby inhibiting apoptosis inflammation, beneficial reducing injury.

An increasing body of evidence indicates that exposure to widespread, environmental and food contaminants such as mycotoxins may cause endocrine disorders infertility. Deoxynivalenol (DON), which is a toxic secondary metabolite produced by Fusarium fungi, can lead multiple harmful effects in humans animals, hepatotoxicity, nephrotoxicity, immunotoxicity, gastrointestinal toxicity, neurotoxicity, genetic toxicity carcinogenicity. Recently, there has been growing concern about DON-induced male Exposure DON its metabolites damage the structure function reproductive organs, resulting impairment gametogenesis thus impaired fertility. Potential molecular mechanisms involve oxidative stress, inflammatory response, mitochondrial dysfunction, apoptosis, cell cycle arrest, pyroptosis, ferroptosis. Moreover, several signaling pathways, including nuclear factor-kappa B, mitogen-activated protein kinase, NLR family pyrin domain containing 3, factor erythroid 2-related 2, AMP-activated apoptotic microRNAs are involved these detrimental biological processes. Research shown antioxidants, small-molecule inhibitors, or proteins (such lactoferrin) supplementation potentially offer protective targeting pathways. This review comprehensively summarizes on mammals, underlying emphasizes potential small molecules therapeutics.
