The transformation of hypoglycin A (HGA), B (HGB), and methylene cyclopropyl glycine (MCPrG) in ruminal fluid batch cultures was investigated, the effect these toxins on culture microorganisms using microbial metabolites measured. An experiment conducted ovine ANKOM RF Gas Production System over four runs, each with an incubation period 48 h. fermenters contained 200 mg (i) a substrate mixture (80% cellulose, 20% starch; CSM), (ii) CSM 1.5 mL solution pure (a 500 ng/mL HGA MCPrG each; PCM), or (iii) 100 sycamore maple seeds (SMS). Each fermenter 30 inoculum (ruminal buffer, 1:2 v/v). For control, autoclaved incubated CSM, PCM, SMS, respectively. Samples were taken from liquid phase analyzed chromatography–tandem mass spectrometry (LC/MS-MS) for metabolites. Microbial activity assessed gas production, short chain fatty acids, NH3 concentration. Additionally, pH redox potentials In concentrations rapidly decreased (p < 0.05), not measurable anymore after 24 h period. initial 4.7 ± 1.4 µg/mL HGA, 19.9 5.41 HGB, 1.2 0.33 MCPrG. increased h, coincidently to decrease HGB concentration 0.05). Summary An 18‐year‐old Saxon Warmblood gelding was referred to an equine clinic in the Czech Republic May 2019 for mild colic. The horse had 7 h of grazing and 30–45 min exercise every day. At clinic, physical examination ruled out gastrointestinal disease, while clinical pathology confirmed rhabdomyolysis (creatine kinase 29,088 IU/L; reference range 110–250 IU/L). Because suspicion atypical myopathy (AM), metabolomic analysis hypoglycin A (HGA), 31 acylcarnitines (ACs) free carnitine performed four dry serum one blood spot. After laboratory confirmation AM patient studied (HGA: 0.757–0.415 μmol/L, ACs elevation), incriminated pasture visited searched source toxin. No sycamore ( Acer pseudoplatanus ) found vicinity pasture. However, several box elder negundo trees were found, toxicological revealed HGA samaras (7.66–296.00 mg/kg), leaves (2.12–9.39 mg/kg) seedlings (339.00 mg/kg). Not only but also tree can be a potential poisoning horses Europe, particularly lowland areas with warm climate.
