Artemisinin, a crucial antimalarial compound synthesized in Artemisia annua, is tightly controlled by various transcription factors. Despite significant progress understanding this regulatory network, further exploration of additional factors needed to improve artemisinin biosynthesis. Here, we identified novel R2-MYB-like factor, AaMYB121, which responds methyl jasmonate (MeJA). Overexpression AaMYB121 transgenic A. annua plants resulted dihydroartemisinic acid (DHAA) content being elevated 2~3 times compared the control, while levels increased 1.4~2 times, significantly boosting Dual-Luciferase (Dual-LUC) assay and yeast one-hybrid (Y1H) analysis demonstrated that directly binds promoter motifs DBR2 (−1146~−1103) ALDH1 (−1558~−1504), thereby triggering transcriptional initiation these genes. Notably, features an elongated DNA-binding domain specifically recognizes AT-rich cis-elements promoters key biosynthetic These findings established as MYB-like factor with strong potential enhance regulation production through targeted modulation, offering valuable approach yields for therapeutic purposes.
