pH and peroxynitrite (ONOO–) are two critical biomarkers to unveil the corresponding status of endoplasmic reticulum (ER) stress mitochondrial dysfunction, which closely related Alzheimer's disease (AD). Simultaneously monitoring ONOO– fluctuations in ER mitochondria during AD progression is pivotal for clarifying interplay between disorders organelles revealing pathogenesis. Herein, we designed synthesized a dual-channel fluorescent probe (DCFP) visualize mitochondria. DCFP possessed excellent sensitivity selectivity without spectral crosstalk was utilized analytes within model cells larval zebrafish. Importantly, could preferentially target normal be enriched after depolarization. With aid DCFP, slower acidification rate than that induced by Aβ oligomers (AβOs) first identified, ascribed relief AβOs-triggered through Ca2+ migration from Moreover, continuous exposure AβOs led overload, accelerating overproduction As result, intracellular oxidative levels were elevated, further exacerbating aggravating turn. The advanced understanding potential this work may offer new insights methodologies studying developed also employed study other diseases dysfunction.

Tendinopathy is one of the most prevalent musculoskeletal disorders, significantly affecting quality life patients. Treatment outcomes can be improved with an early diagnosis and timely targeted interventions. Increasing evidence indicates that ROS endoplasmic reticulum (ER) stress play key roles in modulating differentiation processes tendon-derived stem cells (TDSCs), thereby contributing to initiation progression tendinopathy. However, relationship between ONOO- process, as well various stages tendinopathy, remains unexplored. Herein, we developed two highly specific sensitive fluorescent probes (Rod-Cl Rod-Br) for detecting ER. Rod-Br detect basal levels ER TDSCs measure primary stimulated by interleukin-1β over durations, allowing comparisons chondrogenic osteogenic levels. Additionally, examined variations different tendinopathy treatment rat models vivo discussed potential mechanisms. This research provides a robust tool analyzing dynamics tenogenic TDSCs, offering new insights into pathophysiology
