A rapid and accurate evaluation of a chemical disinfectant’s bactericidal efficacy is crucial for ensuring effective infection control, preventing the spread pathogens, supporting development new disinfectant formulations. In this study, we report rapid, label-free flow cytometry (FCM) protocol evaluating disinfectants. Five commercial disinfectants (alcohols, oxidizing agents, alkylating agents) were evaluated against type strains recommended by EN 13727+A2 ten clinical strains. The FCM method allowed determination through assessment scatter light profiles (FSC-H/SSC-H) count differences. provided results in approximately 4 h showed strong correlation with standard tests (91.4%, sensitivity 0.94 specificity 0.98) that can take up to 48 h. Our represent proof-of-principle reliably assess disinfectants, same day, substantially faster than current growth-based methods. Additionally, study highlights potential detecting occurrence viable but non-culturable bacteria following treatment chlorine-based With its speed, accuracy, capability identify bacterial injuries at single-cell level, powerful tool assessing
