Epigenetic pathways could provide a mechanistic explanation for the inheritance of acquired characteristics, as proposed by Lamarck in 1802, but epigenetic alterations that endow adaptive hereditary traits have rarely been observed. Here, cultivated Asian rice (Oryzasativa L.), we identified an epiallele conferring and heritable cold tolerance, trait enabling northward spread from its tropical origins. We subjected cold-sensitive to multigenerational stress line with stable tolerance. DNA-hypomethylation variation acquiredcoldtolerance 1 (ACT1) promoter region rendered expression insensitive cold. This change is, large part, responsible confirmed DNA-methylation editing. Natural ACT1 DNA hypomethylation is associated tolerance geographic distribution. Hypomethylation at triggers presenting route epigenetic-variation-driven characteristics.
