Exponential scientific data growth presents challenges and opportunities for addressing 56 complex public health issues like the opioid epidemic chronic pain management.57 Despite vast amount of research conducted globally, many datasets remain 58 inaccessible or underutilized due to publication access policies stringent use 59 agreements.The generated through activities is enormous.60 Limited stifles discovery delays translation 61 proven advances into real-world applications.(1)To address these challenges, 62 we argue critical importance open science ecosystems, using National 63 Institutes Health Helping End Addiction Long-term ® Initiative (NIH HEAL 64 ) as a case study.We discuss how building community around can 65 accelerate by enabling dataset integration, increasing statistical 66 power, fostering interdisciplinary collaboration.67 68 Open ecosystems represent fundamental shift in conducted, 69 shared, utilized.An ecosystem comprehensive 70 environment that combines technological infrastructure, standardized protocols, 71 collaborative networks enable transparent sharing integration data, 72 methods, findings.These interconnected integrate collection, 73 storage, processing, analysis, across organizations while adhering FAIR 74 (Findability, Accessibility, Interoperability, Reusability) principles.The 75 encompasses not just technical components sharing, but also human 76 elements: researchers, institutions, funding bodies, partners who work 77 together under shared governance frameworks standards

Genetic studies have identified common and rare variants increasing the risk for neurodevelopmental psychiatric disorders (NPDs). These also been shown to influence structure of cerebral cortex. However, it is unknown whether cortical differences associated with genetic are linked they confer NPDs. To answer this question, we analyzed thickness (CT) surface area (SA) NPDs, in ∼33000 individuals from general population clinical cohorts, as well ENIGMA summary statistics 8 Rare NPDs were preferentially total SA, while mean CT. Larger effects on CT, but not observed NPD medicated subgroups. At regional level, sensorimotor areas, showed higher association areas. We show that schizophrenia- bipolar-disorder- SNPs positive negative effect sizes SA suggesting their aggregated cancel out additive polygenic models. Overall, CT do relate those across individual may be critical non-genetic factors, such medication lived experience disorder.
