Proteins unfold under different environmental insults, among which are heat, cold, high pressure, and chaotropic agents. Understanding the mechanisms that determine unfolding each of these conditions is an important problem directly relates to physical forces three-dimensional structure a protein. Here, we studied residue-specific description transitions marginally stable yeast protein Yfh1 using high-pressure nuclear magnetic resonance. We compared pressure unfolded states demonstrated what has up now been only hypothesis: pressure-unfolded spectrum at room temperature shares features in common with low but not suggesting tighter similarity similar role hydration two processes. By exploring phase diagram mapping onto protein, also show pressure-induced pathways temperatures differ, synergic mechanism between pressure- temperature-induced denaturation.
