Cuproptosis is a novel copper-dependent form of programmed cell death, displaying important regulatory functions in many human diseases, including cancer. However, the relationship between changes mitochondrial viscosity, key factor associated with cellular malfunction, and cuproptosis still unclear. Herein, we prepared phosphorescent iridium (Ir) complex probe for precisely monitoring viscosity during cuprotosis via phosphorescence lifetime imaging. The Ir possessed microsecond lifetimes (up to 1 μs), which could be easily distinguished from autofluorescence improve imaging contrast sensitivity. Benefiting long lifetime, excellent selectivity, targeting abilities, monitor increase (from 46.8 68.9 cP) quantitative manner. Moreover, through situ fluorescence imaging, successfully monitored zebrafish treated lipopolysaccharides or elescolomol-Cu2+, were well-known inducers. We anticipate that this new will useful tool in-depth understanding biological effects cuproptosis.

Dual-state emission (DSE) luminescent materials are a newly discovered category of that exhibit efficient light in multiple states, including dilute solutions, highly concentrated aggregated states and solid states. These effectively address the aggregation-caused quenching (ACQ) observed traditional organic with large conjugated planes, as well limitations aggregation-induced (AIE) materials, which typically do not emit solutions. The design development DSE for organelle imaging applications has attracted considerable interest. In this context, study presents synthesis novel compound, DMSS-AM, characterised by intramolecular hydrogen bonding D-π-A structure. As monophenyl material, DMSS-AM exhibits properties fluorescence quantum yields 22.1% solution 14.0% state. particular, it unique pH-responsive properties, facilitating targeted detection lysosomal pH changes. Confocal laser scanning microscopy cells demonstrated emitters at both low high concentrations affect image quality bio-imaging applications. This advance is expected to significantly broaden applicability future
