Crassulaceae plants are valued for their horticultural, ecological, and economic significance, but genetic improvement is hindered by the absence of efficient stable transformation methods. Therefore, development a tailored method crucial enhancing progress plants. The results indicate that, in experiments conducted on Kalanchoe tetraphylla, K599 strain exhibited highest efficiency (76.67%), while C58C1 was least (21.43%). An acetosyringone concentration 100 μM optimal hairy root transformation, immersion yielded efficiency. Additionally, Silwet L-77 significantly influenced efficiency, with 0.05% leading to decrease. Upon four species, notable differences were observed, K. tetraphylla exhibiting 100%, Sedum alfredii displaying lowest 5%. RUBY reporter gene offers more distinct advantage over GFP observing effects. This study developed simple, feasible, cost-effective obtaining transgenic roots from leaves Crassulaceae. methodology provides technical support function research
