Biofilm formation, which is facilitated by quorum sensing (QS), significantly impairs the performance of pressure-driven membrane systems in water treatment. Herein, we present a quenching (QQ)-phage phoresy system to control biofouling disrupting QS-mediated interactions. This system, composed QQ bacterium Paenarthrobacter nicotinovorans as carriers and hitchhiking lytic phages infecting Pseudomonas aeruginosa with active QS systems, decreased signal levels, inhibited extracellular polymeric substance (EPS), reduced bacterial abundance mature biofilms. Transcriptomic analysis revealed that phage treatment upregulated EPS synthesis genes P. aeruginosa, but bacteria downregulated QS-related genes, weakening secretion antiviral facilitating infect lyse target bacteria. Metabolomic profiling corroborated disrupted pathways critical biofilm stability, including tricarboxylic acid cycle, carbohydrate metabolism, amino metabolism. In off-site cleaning experiments, promoted colonization replaced niche on surface, restored flux (i.e., 90% recovery severely systems). Operation studies showed fouling rates, extended lifespan, maintained salt rejection for reverse osmosis (RO) systems. These findings highlight potential bacterium-phage sustainable alternative conventional chemical treatments damage membranes.
