Potable water reuse has become a key component of sustainability planning in arid regions. Many advanced purification facilities use reverse osmosis (RO) as part treatment, including barrier for microorganisms; however, regrowth after RO treatment been observed. Questions remain about the identity, source, and survival mechanisms microorganisms permeate, but extremely low biomass this is limitation common microbiological methods. Here, we performed high-throughput sequencing on samples collected throughout potable train, by filtering large volumes permeate from membranes during an autopsy. We observed stable, consistent microbial community across three months two parallel trains. contained Burkholderiaceae at high relative abundance, one Aquabacterium sp. that accounted 29% community, average. Like most other microorganisms, sequence was not seen upstream suggest biofilm growing unit process infrastructure, rather than active breakthrough, primary source. A metagenome-assembled genome corresponding to found lack sugar-utilization pathways be able consume molecular weight organic molecules, potentially those pass through RO.
