To fully utilize the Citrus aurantium "Changshanhuyou" resource, hypoglycemic effect and mechanisms of action pectin from Changshanhuyou residue (HYP) were studied. HYP considerably inhibited α-glucosidase, suggesting its potent in vitro activity. In streptozotocin-induced type 2 diabetes mellitus (T2DM) mice, significantly increased body weight, survival rate, hexokinase activity, glycogen content decreased fasting blood glucose, oral glucose tolerance, liver glycated serum protein levels. Furthermore, remarkably improved glycolipid metabolism-related indices both liver, IL-6 TNF-α levels serum, antioxidant enzyme activities liver. also modulated mRNA expression key factors (e.g., Akt, PI3K, IRS2, InsR, GLUT4, G6 Pase, PEPCK, AMPK, GS, GSK-3β) short chain fatty acid production abundance beneficial bacteria. Thus, underlying mechanism may involve activation PI3K/Akt, GS/GSK-3β signaling pathways modulation gut microbiota composition.

Pectin, a class of dietary fiber, has received increasing attention in recent years for its ameliorative effects on metabolic diseases. However, the structural variability pectin leads to differential these The intrinsic mechanism by which pectin, derived from different sources, differentially influences syndrome interacting with gut microbiota and host metabolism remains elusive warrants thorough investigation. To address this, we investigated HG-type pectins apple, citrus, pomelo phenotypic expressions, inflammatory factors, oxidative stress, serum hormone levels mice syndrome. In addition, sought identify pivotal bacterial species metabolites integrating genomics metabolomics approaches. Our exploration also extended relationship between characteristics pectins, microbiota, findings revealed that three diversely improved mice, correlated their beneficial metabolites. Notably, all were closely associated Bacteroides acidifaciens. Besides, potential mediators therapeutic included Bacteroides, Lactococcus, Lachnoclostriclum apple pectin; Colidextribacter, Lachnospiraceae_NK4A136_group, citrus Mucispirillum pectin. Metabolites such as arachidonic acid, kynurenic lithocholic deoxycholic indoleacetic linked microbes, may serve pectin's benefits. Ultimately, molecular weight, degree esterification, monosaccharide composition significantly influenced outcomes. This study contribute more nuanced understanding can inform targeted nutritional strategies modulate management.
