Abstract Aphis gossypii Glover is a serious pest that causes significant economic losses and difficult to control due the rapid development of resistance neonicotinoids. Insect cytochrome P450s play an important role in detoxification insecticides. In this study, we aimed understand function CYP380C12 A. gossypii. The gene was cloned characterized, its relative expression level differed among instars. increased by 48.5% compared with Water group under imidacloprid (IMI) stress for 48 h. Molecular docking predicted binding free energy IMI −7.0 kcal/mol, indicating excellent stability at docked active site, which verified molecular dynamics simulations. Subsequently, silencing efficiency dsCYP380C12 treatment reached 48.9%, 47.0%, 40.0% 24, 48, 72 h, mortality treated 1.45- 1.38-fold dsGFP groups, respectively. Moreover, reproductive period aphids (9.67 ± 0.88) significantly shorter than those sprayed (13.67 1.20). Taken together, these results indicated not only increases sensitivity but also shortens period. Understanding can provide new insights developing innovative strategies

Sitobion miscanthi, the main species of wheat aphids, is one kind harmful pest. Chemical insecticides are important agrochemical products to effectively control aphids. However, broad application has led serious resistance pests several insecticides, and understanding insecticide mechanisms critical for integrated pest management. In this study, SmUGGT1, a new uridine diphosphate (UDP)-glycosyltransferase (UGT) gene, was cloned more strongly expressed in SM-R (the resistant strain imidacloprid) than SM-S susceptible imidacloprid). The increased susceptibility imidacloprid observed after silencing indicating that it can be related imidacloprid. Subsequently, SmUGGT1 regulated post-transcriptionally coding sequences (CDs) by miR-81 verified involved S. miscanthi. This finding crucial roles UGT management pests.
