δ-Tocotrienol, an isomer of vitamin E with anti-inflammatory, neuroprotective and anti-coronary arteriosclerosis properties, is widely used in health care, medicine other fields. Microbial synthesis δ-tocotrienol offers significant advantages over plant extraction chemical methods, including increased efficiency, cost-effectiveness environmental sustainability. However, limited precursor availability low catalytic efficiency key enzymes remain major bottlenecks the biosynthesis δ-tocotrienol. In this study, we assembled complete biosynthetic pathway Yarrowia lipolytica enhanced supply, resulting a titre 102.8 mg/L. The rate-limiting steps was then through various strategies, fusion expression homogentisate phytyltransferaseand tocopherol cyclase, semi-rational design SyHPT multi-copy integration genes. final 5 L bioreactor 466.8 mg/L following fed-batchfermentation. This study represents first successful de novo Y. lipolytica, providing valuable insights into E-related compounds.
