Abstract Multi-modal combination therapy is regarded as a promising approach to cancer treatment. Combining chemotherapy and phototherapy an essential multi-modal endeavor. Ivermectin (IVM) potent antiparasitic agent identified having potential antitumor properties. However, the fact that it induces protective autophagy while killing tumor cells poses challenge its further application. IR780 iodide (IR780) near-infrared (NIR) dye with outstanding photothermal (PTT) photodynamic (PDT) effects. hydrophobicity, instability, low uptake of limit clinical applications. Here, we have structurally modified hydroxychloroquine, inhibitor, synthesize novel compound H780. H780 IVM can form H780-IVM nanoparticles (H-I NPs) via self-assembly. Using hyaluronic acid (HA) modify H-I NPs, nano-delivery system HA/H780-IVM (HA/H-I was synthesized for chemotherapy-phototherapy colorectal (CRC). Under NIR laser irradiation, HA/H-I NPs effectively overcame limitations exhibited cytotoxicity. In vitro in vivo experiment results showed excellent anti-CRC Therefore, our study provides strategy CRC treatment could enhance chemo-phototherapy by modulating autophagy.
