Visualizing and manipulating the behavior of proteins is crucial to understanding physiology cell. Methods biorthogonal protein labeling are important tools attain this goal. In review, we discuss advances in probe technology specific for self-labeling tags, focusing mainly on application HaloTag SNAP-tag systems. We describe latest developments small-molecule probes that enable fluorogenic (no wash) imaging super-resolution fluorescence microscopy. addition, cover several methodologies perturbation or manipulation function towards control biological pathways. Thus, current technical systems means they becoming powerful visualization processes, providing invaluable scientific insights difficult obtain by traditional methodologies. As multiplex tag continues be developed expanded, utility these tags will allow researchers address previously inaccessible questions at forefront biology.
