The transmembrane efficiency of DNA nanomachines greatly affects the performance cell imaging. current methods suffer from long time incubation, low efficiency, and potential cyto-toxicity. Here, we report on a novel gold nanorod nanolayer-mediated photothermal transfection (AuNRs-NMPT) method to deliver nanomachine across membrane. Due good effects, AuNRs nanolayer-functionalized slide (AuNRs-NFS) converted light energy into local heat. Local heat disrupted fluidity membrane for perforation. penetrates rapidly efficiently via those holes under optimal laser power irradiation time. AuNRs-NMPT exhibited 12-fold lower incubation 13-fold higher fluorescent intensity compared that common direct incubation. cells also high viability after being treated with AuNRs-NMPT, due self-repair. efficient, rapid, cytotoxic transfection, ensures sensitive imaging miRNA-21. proposed will promote application clinical cancer diagnosis in future.
