The widespread environmental presence of nanoplastics (NPs) raises significant concerns about their health impacts, particularly on the gastrointestinal system, as NPs are primarily ingested. While previous studies have linked NP-induced intestinal toxicity to oxidative stress and reactive oxygen species (ROS) accumulation, specific mechanisms cell death remain unclear. Here, we showed that environmentally relevant concentrations polystyrene (PS-NPs) induced ferroptosis, a form lipid peroxidation-driven death, in epithelial cells. Using epithelial-specific Nrf2-deficient mice (Nrf2fl/fl-VilCre+) human Caco-2 cells, demonstrated Nrf2, key regulator, play protective role against PS-NP-induced ferroptosis. PS-NP exposure disrupted ether phospholipid metabolism, leading accumulation polyunsaturated fatty acid-ether phospholipids heightened peroxidation intestines Nrf2fl/fl-VilCre+ mice. This increased susceptibility cells Additionally, high-fat diet further exacerbated this effect, suggesting individuals with reduced NRF2 activity poor dietary habits may be especially vulnerable damage. Our findings offered new insights into molecular underscored risks posed by exposure, populations compromised antioxidant defenses.

Background: Research evidence has recently shown an association between autism spectrum disorder (ASD) and inflammation. For example, the expression of inflammatory cytokines is abnormal in children with ASD, maternal inflammation can lead to ASD-like behavior offspring. These studies suggest that plays important role occurrence development ASD. Inflammatory may, therefore, be potential biomarkers for In present study, we sought systematically identify Methods: We used Olink proteomics comprehensively examine differentially expressed inflammation-related proteins 60 ASD 28 typical (TD). validated our findings using published data. Results: A total 18 were TD groups. Compared group, all was up-regulated group. Furthermore, eight showed good diagnostic efficacy, as delineated by area under curve (AUC) values > 0.7. To knowledge, this first time interleukin-17C (IL-17C), chemokine ligand (CCL)-19, CCL20 have been detected plasma (with AUC 0.839, 0.763, 0.756, respectively). also found there a negative correlation SRS scores. Conclusions: Multiple markers increased IL-17C, CCL19, exhibit biomarker candidates Elevated levels may enhance social ability

As an emerging contaminant, nanoplastics have evolved into a global ecological issue. Studies shown that induce neurotoxicity across species, however, the causal mechanism remains unknown. This study aimed to explore underlying caused by polystyrene (PS-NPs) via microbiota-gut-brain axis in immature mice, which serve as model of infants and young children who are at higher exposure risk NPs. The results indicated while only minority PS-NPs reached brain after exposure, they still had significant neurotoxic effects, reflected abnormalities behavior, biochemical marker levels histopathology. Proteomics quantification analyses revealed proteostasis imbalance mediated lysosomal proteasome dysfunction is key reason for induced neurotoxicity. Further, we confirmed indirect role gut microbiota through 16S rDNA fecal transplantation. Crucial bacterial species such Eubacterium coprostanoligenes potentially act indicators dysbiosis exposure. Notably, first estimated effect on attributed mice 39.20% high-dimensional mediation analysis. Trehalose was identified mediator connecting brain, crucial trehalose supplementation highlighted remodeling alleviate mice. These findings expected contribute deeper understanding assessment health protection nervous system from early life.
