Nanopore sensors have been used as ultrasensitive tools for single-particle detection based on ionic current measurement. This simple, yet powerful technique allows researchers to acquire various physical properties of individual particles in a label-free manner. mini-review describes the recent progress nanopore technology demonstrated by our group. We first focus major advancements architecture contributing high-spatial resolution, followed strategy designed long-term analysis. Then, we summarize application infection diagnosis using machine learning. Following that, discuss its potential gene therapy, facilitated high spatial resolution. Furthermore, also highlighted applications next-generation that contribute healthier future.

Melittin, a natural peptide found in bees, has been shown to induce pore formation cell membranes. However, its artificial mutant, MelP5 can do so at concentrations 200 times lower than melittin. The mechanism of the enhanced portion ability is not fully understood. By conducting all-atom molecular dynamics (MD) simulations, we that forms stable macro-molecular sized. Our results suggest mutation five amino acids from melittin reduces electrostatic repulsion between peptides and strengthens hydrophobic interactions lipid tails, resulting larger pore. Furthermore, cholesterol (CHOL), which occupies 30% mammalian membranes, plays crucial role enhancing MelP5. As amount CHOL increases, becomes larger, more stable, quickly. presence also promotes oligomers, further support findings indicate insertion into membrane surface state peptides, thereby stabilizing These highlight important permeabilization by provide new insights action membrane-active antimicrobial peptides.
