Superparamagnetic iron oxide nanoparticles (SPIONs) exhibit unique properties for diverse biomedical applications, including drug delivery and diagnostic imaging. Actively targeted SPIONs enhance to diseased sites, reducing side effects enhancing treatment efficacy. However, the development of reproducible functionalization protocols is challenged by erratic behavior in suspensions, such as agglomeration sedimentation. In this study, a method developed systematically optimized attach Fc-region antibodies onto silica-coated via click chemistry, ensuring controlled ligand orientation on particle surface. The synthesis successive modifications with organic moieties are presented resulting final conjugation targeting intercellular adhesion molecule 1 (ICAM1). This protein upregulated epithelial cell surfaces during gastrointestinal inflammation. Thermogravimetric analysis infrared spectroscopy confirm successful SPION after each modification step.
