Pathogenic platelet factor 4 (PF4) antibodies contributed to the abnormal coagulation profiles in COVID-19 and vaccinated patients. However, mechanism of what triggers body produce these has not yet been clarified. Similar patterns many comparable features between virus heparin-induced thrombocytopenia (HIT) have reported. Previously, we identified a new autoimmunity HIT which PF4-antibodies self-clustered PF4 exposed binding epitopes for other pathogenic PF4/eparin antibodies. Here, first proved that SARS-CoV-2 spike protein (SP) also binds PF4. The was evidenced by increase mass optical intensity as observed through quartz crystal microbalance immunosorbent assay, while switching surface zeta potential caused interactions affinity PF4-SP were evaluated dynamic light scattering isothermal spectral shift analysis. Based on our results, proposed generation We further validated changes interaction SP found their differs from ACE2–SP binding.
